A contribution to our knowledge of coca leaf content substances

Abstract

Dried leaves were crushed and extracted in ethanol. The alkaloid fraction was shaken out of the extract. Then it was divided on a silicaged filled column. Partial fractions were purified by further column chromatography and preparative thin-layer chromatography. Identification of single alkaloids was made by comparison with standards on TLG, measurement of IR, UV, NMR and MS spectra and comparison of retention times with standards by gas chromatography. In this way cocaine, trans and cis-methylcinnamoylecgonine, ecgonine methylester, benzoylecgonine, pseudoecgonine methylester, cuscohygrine and mesodihydrocuscohygrine were identified. In the case of meso-di-hydrocuscohygrin, which is a quite new alkaloid isolated from coca leaves, comparison of NMR-spectra after its semisynthetic preparation from a mixture of isolated dl- and meso-cuscohygrine was used for its identification and stereochemical definition.
