Abstract

Kinetic resonance Raman spectroscopy as a function of pH has been utilized to determine the pK of Schiff base deprotonation during the bacteriorhodopsin photochemical cycle. It is shown that the pK of Schiff base deprotonation is between 9.9 and 10.3, microseconds after light absorption and is >12 before photon initiation of photochemical cycling associated with proton pumping.
